If you run up to a closed door (which then opens because you are next to it) can you then attack someone adjacent or non adjacent (with line of sight) in the same turn if you moved six squares or less to get to the door in the same move?

Second, when counting to see if an enemy is within six squares are they considered to be within six squares for purposes of non adjacent attacks or force attacks/abilities if the sixth spot is an adjacent square that is diagonal to the intended targeted enemy?

Do you count the square you are starting in and occupying when determining if an enemy/potential target is six squares away.

No, because the door opens at the end of your turn.

I'm not sure that I understand the question. Are you asking if you use Grenades (or another similar ability) on an enemy within 6 squares, but an adjacent enemy is 7 squares away, would it damage that enemy? If that is what you are asking, then the answer is yes, because it is the target that has to be within 6 squares. If you are asking something else then I misunderstood and I have no idea. Confused
